Yesterday before end work was working on microbiological test on Corn Cob, and took pics with my favorite machine, which is also xuan's favorite: AUTOCLAVE! So let me take u thru the steps of doing a microbiological experiment:
The product that we are testing:

1st u must have the agar powder, for this we are using a general agar to grow all kinds of bacteria so here it is:

then, u must weigh according to the specs, for this case is 2.6g


Following, u must pour water to make it into a solution, rmb to use distill water..

Bring the solution to heat:

In the mean time, prepare the test tube filled with water, as u want a sterile water when doing ur experiment, to prevent contamination:

And also a number of pipette tips:

The solution have boiled :

The petri dishes are sterile, so u need not sterilise it:

This is the autoclave machine, it looks so alien, haha:

Add water in it, and place the items in the 2 tub in it, close it tightly:

The temperature and pressure is set:

The temperature will rise:

Keep rising:

And continue to rise:

Still rising:

It will go no higher than this, as there is a sensor, due to the pressure water boils at 121degree instead of 100 degree:

Leave it for 15 - 20 mins, at the mean time, on ur UV light at the laminar flow hood:

After that, pour ur agar on the plates allow it overnight, following day, set ur equipments in ur fumehood, make sure there is no contamination:

Do your serial dilution, and dun forget to vortex it to ensure homogeneously mixture:

After that, pipette the serial dulution onto your agar plates:


Rmb to soak ur spreader in ethanol, and set it on fire before usage, this is also to ensure sterile conditions:

Spread the liquid on the agar plates, please be gentle, or u will crack it...and dun forget to label them:
